| Synthesis | General procedure for the synthesis of ethyl 3-cyano-1H-pyrrole-2-carboxylate from the compound (CAS:147299-51-2): a solution of 0.5 M ammonia (NH3) in dioxane (22 ml) was added slowly and dropwise to a solution of ethyl acetate (EtOAc, 30 ml) of 1b (1.62 g) at 60 °C. The reaction mixture was heated to reflux for 18 hours. Upon completion of the reaction, the mixture was cooled to room temperature and the solvent was subsequently removed by rotary evaporation. The residue was purified by silica gel column chromatography to give the final light yellow solid product 88a (0.37 g). | | References | [1] Patent: WO2016/7185, 2016, A1.
