| Synthesis | To a solution of tetrahydrofuran (THF, 6 mL) containing 2,4-dichloro-5-fluoro-6-methylpyrimidine (563 mg, 3.11 mmol), a methanolic solution of 25% sodium methanolate (671 mg) was slowly added under cooling at 0°C. The reaction mixture was slowly warmed up to room temperature over 1 hour. After completion of the reaction, the reaction solution was diluted with water and extracted with ethyl acetate (EtOAc). The organic phases were combined, concentrated and the residue was purified by silica gel column chromatography (eluent: ethyl acetate/hexane) to afford 2-chloro-5-fluoro-4-methoxy-6-methylpyrimidine in quantitative yield. | | References | [1] Patent: WO2009/131974, 2009, A1.
