| Uses | Ethyl Cytosine-5-carboxylate is a reagent for the synthesis of cytidine-5-carboxylic acid, which are potential anticancer agents. | | Synthesis | General procedure for the synthesis of ethyl 2-hydroxy-4-aminopyrimidine-5-carboxylate from RSSYY (lurasidone hydrochloride)-16: 400 ml of diethyl ether and 500 ml of tetrahydrofuran were added to a 2500 ml round bottomed flask, followed by the addition of 30 ml of sodium tert-butanol to synthesize intermediate 2. After stirring the mixture, 183 g (1 mol) of intermediate 1 and (0.1 mol) of sodium magnesium oxide nanopowder. The reaction mixture was heated to 75°C and maintained at this temperature for 2.5 hours. Upon completion of the reaction, it was quenched, cooled to room temperature and filtered to collect the yellow solid product. The resulting solid was dissolved in 500 ml of water with continuous stirring and gradual addition of water until the yellow solid was completely dissolved. The pH of the solution was adjusted to 6~7 with dilute hydrochloric acid (prepared from concentrated hydrochloric acid and water at a volume ratio of 1:3), at which time a white solid precipitated. The white solid was collected by filtration and washed with a small amount of acetone.
