步骤1. 在0℃冰浴条件下,向搅拌的3-羟基哌啶-1-羧酸叔丁酯(5g,24.84mmol)和三乙胺(5.027g,6.924mL,49.68mmol)的二氯甲烷(50mL)溶液中缓慢加入甲基磺酰氯(3.130g,2.115mL,27.32mmol)。反应混合物在3小时内逐渐升温至室温。反应完成后,用水和1M盐酸洗涤反应混合物,随后用硫酸镁干燥,过滤并真空浓缩,得到3-((甲基磺酰基)氧基)哌啶-1-羧酸叔丁酯,为黄色油状物(7.9g,定量收率,含少量残留溶剂)。1H NMR(400.0MHz,CDCl3)δ1.49(s,9H),1.60-1.57(m,1H),2.04-1.79(m,3H),3.08(s,3H),3.38-3.32(m,1H),3.50-3.43(m,1H),3.70-3.60(m,2H)和4.74(br s,1H)ppm。
