Uses5-Chloro-1,2,3,4-tetrahydro-2-(phenylmethyl)-2,6-naphthyridine is an intermediate used to prepare substituted naphthyridines as FAAH modulators useful in prevention and treatment of FAAH-mediated diseases.
SynthesisA round bottom flask was charged with 6-benzyl-5,6,7,8-tetrahydro-2,6-naphthyridin-1-ol (7 g, 29 mmol) and phosphorochloridic acid {LeonidChemicals, India; add 80 mL) and heated to reflux for 16 hours to monitor the progress of the reaction by TLC. Upon completion of the reaction, POCl3 was distilled and the residue was quenched with ice-cold water and then extracted with dichloromethane (3 100 mL). The combined organic layers were dried with anhydrous sodium sulfate and the solvent was removed under reduced pressure. The residue was purified by column chromatography (methanol/dichloromethane, 5:95) to afford the desired product 2-phenylmethyl-5-chloro-1,2,3,4-tetrahydro-[2,6]diazanaphthalene (80% yield).
