7,8-二溴-2-氯-9-甲基-5,6-二氢-4H-咪唑并[4,5,1-IJ]喹啉的合成步骤如下(方法2A):将2-氯-9-甲基-5,6-二氢-4H-咪唑并[4,5,1-IJ]喹啉(11.7 g,56 mmol,方法14A)溶解于预先冷却至0℃的乙腈(250 mL)中。随后,分批加入N-溴代琥珀酰亚胺(50.0 g,282 mmol),加料时间超过1小时。反应混合物在0℃下继续搅拌30分钟,之后升至室温并搅拌48小时。反应完成后,通过减压蒸馏去除溶剂。将残余物溶解于二氯甲烷中,并用饱和碳酸氢钠水溶液洗涤。有机层经无水硫酸钠干燥后,过滤并浓缩滤液,得到18.6 g目标产物7,8-二溴-2-氯-9-甲基-5,6-二氢-4H-咪唑并[4,5,1-IJ]喹啉,收率为90%。产物通过LC-MS(m/z 364.8 [M + 1])和1H NMR(600 MHz, DMSO)δ 4.12(t, 2H, CH2),2.88(t, 2H, CH2),2.57(s, 3H, CH3),2.22-2.18(m, 2H, CH2)进行表征。
