| | ATTO 647N NHS ester Usage And Synthesis |
| Description | ATT 647N NHS ester is an amine-reactive dye for labeling various amine-containing molecules in an aqueous phase without using any organic co-solvent. This product is beneficial for the labeling of peptides and proteins that denature in the presence of organic co-solvents, as well as for proteins with low solubility.
ATT 647N is a rhodamine-based far-red fluorophore with strong molar absorption, high fluorescence quantum yield, and excellent thermal and photostability. ATT 647N fluorescence is independent of pH in the range of 2 to 11, which supports its application under diverse experimental conditions.
Unlike cyanine dyes, ATT 647N exhibits enhanced resistance to atmospheric ozone degradation, making it highly suitable for microarray and other high-precision applications such as single-molecule detection, super-resolution microscopy techniques (e.g., SIM and STED), flow cytometry (FACS), and fluorescence in situ hybridization (FISH). | | Uses | Atto 647N belongs to a new generation of fluorescent labels for the red spectral region. The dye is designed for application in the area of life science, e.g. labeling of DNA, RNA or proteins. Characteristic features of the label are strong absorption, excellent fluorescence quantum yield, high photostability, excellent ozone resistance, good solubility, and very little triplet formation. Atto 647N is a cationic dye. The active ester of Atto 647N fluorescence dye reacts with amino groups under mild conditions. After coupling to a substrate the dye carries a net electrical charge of +1. In common with most Atto-labels, absorption and fluorescence are independent of pH in the range of 2 to 11, used in typical applications. As supplied Atto 647N consists of a mixture of two isomers with practically identical absorption and fluorescence properties. | | Abs/Em Maxima | 644/662 nm | | Fluoresene quantum yield | 0.68 | | Extinction Coefficient | 105000 | | CF260 | 0.08 | | CF280 | 0.05 |
