| Uses | Zymosan A from Saccharomyces cerevisiae has been used:
- as a positive control to determine the activation of complement cascade
- for respiratory burst (RB) assay
- to assess its effects on performance, immunity, heat shock protein and apoptosis related gene expressions of dairy cows under heat stress (HS)
- a stimulating agent in Krebs-Ringer phosphate (KRP) medium
- to induce thermal hyperalgesia in mice
| | Biochem/physiol Actions | Zymosan is prepared from yeast cell wall and consists of protein-carbohydrate complexes. It is used to induce experimental sterile inflammation. In macrophages, zymosan-induced responses include the induction of proinflammatory cytokines, arachidonate mobilization, protein phosphorylation, and inositol phosphate formation. Zymosan A also raises cyclin D2 levels suggesting a role for the latter in macrophage activation besides proliferation. It potentiates acute liver damage after galactosamine injection suggesting that certain types of nonparenchymal cells other than Kupffer cells are involved in zymosan action. Zymosan A, a 13 beta glucan contributes to the development of organic dust-induced respiratory disease. | | in vivo | Zymosan A (25.0mg/kg; IP at 12 and 2h before IR) mitigates IR-induced intestinal injury in mice[1]. Zymosan A can be used in animal modeling to create peritonitis models.
Induction of peritonitis[2] Background
Zymosan A can mediate the release of pro-inflammatory cytokines including IL-1β, TNF-α, and HMGB1, inducing an inflammatory response leading to peritonitis.
Specific Modeling Methods
Mice: C57BL/6 male 3-7 months old Administration: 100 or 500 mg/kg i.p.

(1) ChAT-eGFP mice were randomly divided into two groups, and baseline rectal temperature and electrocardiogram (ECG) were measured in each group before injection.
(2) Preparation of Zymosan A Suspension: Weigh Zymosan A and sterilise for 30 minutes with UV light, add sterile filtered saline and vortex thoroughly, store at room temperature overnight and vortex again before use to ensure thorough mixing.
(3) Rectal temperature and ECG were recorded hourly 4 and 24 hours after Zymosan A injection, respectively.
Modeling Indicators Phenotypic observation: Hypothermia; cardiac insufficiency, including increased variability of baseline heart rate (HR), moderate slowing of ventricular conduction, and decreased cardiac contractility; and bacterial translocation into the peritoneal cavity and bloodstream.
