| Synthesis | The general procedure for the synthesis of 2-amino-3-methoxypyrazine from 2-amino-3-chloropyrazine is as follows:
Pre-step A: Synthesis of 3-methoxy-2-pyrazinamine
3-Chloro-2-pyrazinamine (200 mg, 1.544 mmol) and sodium methanol (250 mg, 4.63 mmol) were dissolved in methanol (3.9 mL) and the mixture was placed in a microwave reactor, heated to 130°C, and reacted for 60 min. Upon completion of the reaction, the crude product was purified by reversed-phase high performance liquid chromatography (RP-HPLC) to afford 3-methoxy-2-pyrazinamine (113 mg, 0.903 mmol, 59% yield).
