In a 250 mL three-necked flask, under nitrogen atmosphere, 0.04 mol 2-iodo-3-nitronaphthalene (Intermediate 2-1) and 0.08 mol SnCl2 were added and dissolved in 100 mL of ethanol. The mixture was heated to reflux and reacted for 6 hours. The progress of the reaction was monitored by thin layer chromatography (TLC) and the reaction was stopped after confirming the complete consumption of intermediate 2-1. The reaction solution was allowed to cool to room temperature and filtered. The filtrate was rotary evaporated under reduced pressure until no fraction evaporated. Finally, the intermediate 2-2 (3-iodonaphthalen-2-amine) was purified by neutral silica gel column chromatography and analyzed by high performance liquid chromatography (HPLC) with 99.1% purity and 75.9% yield.
