| Application | 2-Acetylpyrene can be used as a pharmaceutical synthesis intermediate. | | Synthesis | The synthesis of 2-Acetylpyrene method is as follows: Stir a solution of 2-acetyl-4,5,9,10-tetrahydropyrene (2.01 mmol) and 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (2.1 equivalents, 0.96 g, 4.23 mmol) in dry THF (25 mL) at reflux (80 °C) under a stream of N2 for 2 days. Remove the solvent by rotary evaporation under reduced pressure. Wash the remaining residue with 5% aqueous KOH (15 mL). Extract the residue with DCM (3 × 30 mL). Dry the combined extracts over MgSO4. Filter the residue. Evaporate the filtrate to dryness. Purify the product by column chromatography (silica gel, ethyl acetate:n-hexane 1:9). Crystallize the crude product from EtOH.1 | | References |
