FAP分子背景
FAP（也称为seprase）是一种II型跨膜丝氨酸蛋白酶。质膜和可溶性形式都表现出脯氨酸切割后的内肽酶活性，对Ala/Ser-Gly-Pro-Ser/Asn/Ala共有序列有明显的偏好。也降解明胶，热变性I型胶原蛋白。还具有二肽基肽酶活性，优选Ala-Pro、Ile-Pro、Gly-Pro、Arg-Pro和Pro-Pro。质膜形式与DPP4、PLAUR或整合素结合，参与细胞外基质（ECM）的细胞周蛋白水解，从而促进细胞通过ECM的粘附、迁移和侵袭。通过降解短链蛋白多糖促进胶质瘤细胞通过脑实质的侵袭。通过以丝氨酸蛋白酶活性无关的方式调节细胞增殖和存活，在黑素细胞中充当肿瘤抑制因子。
