Immunohistochemistry was performed on human pancreatic ductal carcinoma and human appendix tissue. High-pressure and temperature Sodium Citrate pH 6.0 was used for antigen retrieval. Following antigen retrieval, tissues were blocked in a 10% goat serum in wash buffer solution for 30 minutes at room temperature and endogenous peroxidase activity quenched with Peroxidase Suppressor. Tissue was then probed with a Recombinant Synaptophysin Antibody (Ab129952) at a dilution of 1:100 in 10% goat serum for overnight at 4℃. Using a Goat Anti-Mouse IgG H&L (Biotin) (Ab156253) as secondary antibody, and detection was performed using a Streptavidin (HRP) (np156148) followed by colorimetric detection using DAB .Tissues were then counterstained with hematoxylin and prepped for mounting and imaging.

3 μg/lane of Recombinant Synaptophysin Antibody (Ab129952) was resolved with SDS-PAGE under reducing (R) and non-reducing (N) conditions and visualized by Coomassie® Blue staining, showing the band at 27.0 kDa (Light Chain) & 53.2 kDa (Heavy Chain) under reducing conditions and 173.2 kDa under non-reducing conditions.
