G4S linker是单链抗体最常见的连接肽。它由重复序列（Gly-Gly-Gly-Gly-Ser）组成，通常在scFv片段中以15-mer（G4S）3或20-mer（G4S）4的形式存在。G4S linker已成为绝大多数CAR不可或缺的一部分，因为它连接scFvs的VH和VL识别结构域。G4S linker通常用于靶向实体瘤抗原（如HER2和CEA）的CAR，而惠特洛/218接头肽主要用于靶向血液系统恶性肿瘤（如CD19 CAR产物）的CAR构建。
