背景(Background)
酪氨酸蛋白激酶Mer(MERTK)是TYRO3/AXL/Mer(TAM)受体激酶家族的成员,编码一种跨膜蛋白,具有两个纤维连接蛋白III型结构域、两个Ig样C2型(免疫球蛋白样)结构域和一个酪氨酸激酶结构域。在配体激活后,与GRB2或PLCG2相互作用,诱导MAPK1、MAPK2、FAK/PTK2或RAC1的磷酸化。MERTK信号在巨噬细胞清除凋亡细胞、血小板聚集、细胞骨架重组和吞噬等各种过程中起着重要作用。视网膜色素上皮(RPE)作为杆状外节片段吞噬作用的调节因子。通过激活STAT1,在抑制Toll样受体(TLRs)介导的先天免疫反应中也起着重要作用,STAT1选择性诱导细胞因子信号SOCS1和SOCS3抑制剂的产生。
