描述	MOG (35-55) mouse, rat is a minor component of the central nervous system myelin with high brain specificity and strong immunogenicity. It can induce T and B cell responses, trigger relapsing-remitting neurological disorders with plaque-like demyelination, and is commonly used for inducing experimental autoimmune encephalomyelitis (EAE) models.
动物实验	Female C57BL/6 mice (n?=?20) were induced by myelin oligodendrocyte glycoprotein (MOG)35-55 peptide.?At 14 days after immunization, T cells were isolated from the spleen and purified as CD4 and CD8 T cells by using CD4 and CD8 isolation kits, and then the purity was determined by flow cytometric analysis.?These cells were stimulated by MOG35-55 peptide and applied to proliferation assays.?The interferon-gamma (IFN-γ) and interleukin (IL)-4 secretion of supernatant of cultured CD4 and CD8 T cells were measured by enzyme-linked immunosorbent assays (ELISA).?For adoptive transfer, recipient mice were injected with MOG35-55-specific CD8 or CD4 T cells.?EAE clinical course was measured by EAE score at 0-5 scale and spinal cord was examined by staining with hematoxylin and eosin and Luxol fast blue staining[1].
体外活性	方法：树突状细胞（DC）上样 MOG35-55 肽，以 10 μg/mL 的浓度添加到培养物中。2小时后，将脂多糖（LPS，Sigma-Aldrich）以0.25μg/ mL的浓度加入培养物中,两天后（培养的第 8 天），使用流式细胞术对 DC 进行表型分析。结果：使用 Myelin Oligodendrocyte Glycoprotein Peptide (35-55), mouse, rat (MOG (35-55))的培养物在MOG35–55的DC培养物中，分泌的细胞因子为IL-6、IL-10、IFN-γ、TNF-α和TGF-β，TGF-β浓度最高;IL-4 和 IL-17 在任何培养物中均未检测到。[2]
体内活性	方法：雌性 C57BL/6 小鼠（n = 20）皮下注射 200 μL 含有 200 μg 的Myelin Oligodendrocyte Glycoprotein Peptide (35-55), mouse, rat (MOG (35-55))，免疫后14 天，从脾脏中分离出T细胞，使用CD4和CD8分离试剂盒纯化为CD4和CD8 T细胞，然后通过流式细胞术分析测定纯度;这些细胞受到Myelin Oligodendrocyte Glycoprotein Peptide (35-55), mouse, rat (MOG (35-55))刺激并应用于增殖测定。采用酶联免疫吸附试验（ELISA）检测培养的CD4和CD8 T细胞上清液的干扰素-γ（IFN-γ）和白细胞介素（IL)-4分泌。结果：CD8/CD4 富集后，CD8CD3 和 CD4CD3 细胞分别占总 CD3 细胞的 86% 和 94%。与CD4 T细胞相比，CD8 T细胞分泌的IFN-γ和IL-4较少。[1]
