ATG12 antibodies are essential tools for studying the molecular mechanisms of autophagy, a conserved cellular process responsible for degrading damaged organelles and proteins. Autophagy-related gene 12 (ATG12) is a ubiquitin-like protein critical for autophagosome formation. During autophagy initiation, ATG12 undergoes a conjugation process: it is first activated by the E1-like enzyme ATG7. transferred to the E2-like enzyme ATG10. and finally covalently linked to ATG5. The ATG12-ATG5 conjugate interacts with ATG16L1 to form a complex that facilitates the lipidation of LC3 (microtubule-associated protein 1A/1B-light chain 3), a key step in autophagosome membrane expansion.

ATG12 antibodies are widely used in research to detect ATG12 expression levels, conjugation status, and subcellular localization via techniques like Western blotting, immunofluorescence, and immunoprecipitation. These antibodies help elucidate autophagy dysregulation in diseases such as cancer, neurodegenerative disorders, and infections. For example, reduced ATG12-ATG5 conjugation may indicate impaired autophagy, linked to pathological protein aggregation in Alzheimer’s disease. Both monoclonal and polyclonal ATG12 antibodies are available, with specificity validated across species (human, mouse, rat). Researchers also use them to assess autophagy flux when combined with other markers (e.g., LC3-II/p62). Understanding ATG12’s role through antibody-based assays contributes to therapeutic strategies targeting autophagy pathways.
