该基因编码的蛋白——低密度脂蛋白相关蛋白2（LRP2/megalin）——是一种多配体内吞受体，广泛表达于多种组织，但主要存在于肾脏等吸收性上皮组织。该糖蛋白具有巨大的氨基端胞外结构域、单个跨膜结构域及较短的羧基端胞质尾。其胞外配体结合域可结合多种大分子，包括白蛋白、载脂蛋白B/E及脂蛋白脂肪酶。
LRP2蛋白对脂蛋白、固醇、维生素结合蛋白及激素等配体的重摄取至关重要，同时参与细胞信号传导：胞外配体包括甲状旁腺激素与形态发生素音猬因子（sonic hedgehog），胞质配体则含MAP激酶支架蛋白和JNK互作蛋白。该膜受体的循环利用受其胞质域磷酸化调控。此基因突变可导致Donnai-Barrow综合征（DBS）及眼-耳-肾综合征（FOAR）。
