β-连环蛋白（CTNNB1）是经典Wnt信号通路的核心下游组分。无Wnt信号时，其与AXIN1、AXIN2、APC、CSNK1A1及GSK3B形成复合物，通过BTRC介导CTNNB1蛋白N端丝氨酸/苏氨酸残基磷酸化及泛素化修饰，进而被蛋白酶体降解。当Wnt配体存在时，CTNNB1避免泛素化降解并在细胞核内累积，作为TCF/LEF家族转录因子的共激活因子，启动Wnt应答基因表达。该蛋白还参与以下生物学过程：
1、细胞粘附调控：作为E-钙黏蛋白/连环蛋白粘附复合物组分
2、中心体解聚：负向调控中心体黏连
3、胰岛素内化：参与CDK2/PTPN6/CTNNB1/CEACAM1信号轴
4、肿瘤恶性化：阻断肾脏/肠道上皮细胞失巢凋亡，通过下调DAPK2促进锚定非依赖性生长
5、核体形成抑制：阻碍PML核体形成，抑制RANBP2介导的PML小泛素化修饰
6、神经发生调控：维持交感神经母细胞处于细胞周期内
7、软骨分化调节：与SOX9相互作用竞争性占据TCF/LEF结合位点，从而抑制Wnt信号通路介导的软骨细胞分化
