前列腺特异性膜抗原（PSMA），亦称叶酸水解酶1（FOLH1）、谷氨酸羧肽酶2（GCP2）或N-乙酰化α-连接酸性二肽酶I（NAALAD1），隶属肽酶M28家族M28B亚家族。该蛋白在pH＞6.5环境中稳定存在，是一种II型跨膜锌金属肽酶，在神经系统、前列腺、肾脏及小肠中呈现高表达。其以同源二聚体形式存在，每个亚基结合2个锌离子，为NAALAD酶活性所必需。PSMA的催化机制涉及释放未取代的C端谷氨酰残基，典型作用底物为乙酰天冬氨酸-谷氨酸（Ac-Asp-Glu）或叶酰聚-γ-谷氨酸。该酶兼具叶酸水解酶与N-乙酰化α-连接酸性二肽酶（NAALAD酶）活性，并对三聚α-谷氨酸肽具有底物偏好性。GCP-2/PSMA参与前列腺肿瘤进展过程，同时表现二肽基肽酶IV型活性，在体外实验中可特异性切割甘氨酸-脯氨酸-AMC（Gly-Pro-AMC）。
