背景介绍
酪氨酸蛋白激酶跨膜受体ROR1（亦称神经营养酪氨酸激酶受体相关蛋白1，NTRKR1）属于蛋白激酶超家族、酪氨酸蛋白激酶家族及ROR亚家族成员。该受体包含1个FZ（卷曲）结构域、1个Ig样C2型结构域、1个Kringle结构域和1个蛋白激酶结构域。ROR1在胚胎发育早期高表达，至第16天左右表达水平急剧下降，在成年组织中仅维持极低表达水平。其短亚型（Isoform Short）在胎儿及成人中枢神经系统以及多种人类癌症（包括源自中枢神经系统或周围神经系统神经外胚层的肿瘤）中显著表达。ROR1可通过与ε型酪蛋白激酶1（CK1ε）相互作用，激活磷酸肌醇3-激酶介导的AKT磷酸化及cAMP应答元件结合蛋白（CREB）信号通路，从而促进肿瘤细胞生长。
