TDP-43，全称为反式激活应答（TAR）-DNA结合蛋白，分子量为43 kDa，由位于1号染色体的TARDBP基因编码。它是一种RNA/DNA结合蛋白，结构上类似于典型的核内不均一核糖核蛋白（hnRNP）家族成员，参与转录调控和RNA加工。该蛋白与散发性和家族性肌萎缩侧索硬化症以及额颞叶痴呆的发病密切相关。TDP-43主要定位于细胞核内，但在病理条件下会转位至细胞质。其胞质聚集、磷酸化、泛素化及截断现象是TDP-43蛋白病的核心病理特征。
