Anumigilimab (CSL-324) is a fully human monoclonal antibody of the IgG4 isotype that functions as an antagonist targeting the Granulocyte Colony-Stimulating Factor Receptor (G-CSFR). By blocking this receptor, Anumigilimab inhibits the signaling pathway triggered by its ligand, G-CSF. This action is designed to reduce G-CSF-mediated neutrophilia and neutrophil infiltration into inflamed tissues without causing systemic neutropenia (a deficiency in neutrophils). The IgG4 isotype minimizes the risk of target cell depletion via effector functions. Anumigilimab has been investigated as a therapeutic agent for various chronic inflammatory and autoimmune diseases characterized by excessive neutrophil activity and G-CSF dysregulation, such as Hidradenitis Suppurativa.
