Manelimab is a human IgG1-lambda monoclonal antibody that specifically targets programmed death-ligand 1 (PD-L1, CD274), an immune checkpoint protein. As an IgG1 isotype, it is engineered to potentially engage immune effector functions such as antibody-dependent cellular cytotoxicity (ADCC) in addition to checkpoint blockade. By binding to PD-L1 with high affinity, it blocks the interaction between PD-L1 and its receptor PD-1 on T cells. This inhibition reverses the suppression of T-cell activity, thereby restoring anti-tumor immune responses. It is being developed as an immune checkpoint inhibitor for the treatment of advanced solid tumors.
