CREBide is a synthetic 13 amino acid peptide that has been reported as a PKA substrate. Delta CREB is a splicing variant of cAMP response element binding protein (CREB). A synthetic peptide based on delta CREB phosphorylation sequence. Delta CREB and CREBide were tested as substrates for cAMP dependent protein kinase (cAK). The apparent Km of cAK for CREBtide phosphorylation is 3.9 μ M, which is 10 times lower than the synthetic peptide substrate Kemptide (Km=39 μ M) most commonly used for cAK measurement. The Vmax value of CREBtide is 12.4 mumol/(min. mg), and the Vmax value of Kemptide is 9.8 mumol/(min. mg). The apparent Km of cGMP dependent protein kinase (cGK) for CREBtide phosphorylation is 2.9 μ M, with a Vmax value of 3.2 mumol/(min. mg). Compared to cAK, delta CREB and CREBide are phosphorylated by cGK at a much slower rate, indicating that the high cAK/cGK specificity exhibited by delta CREB is preserved by the peptide
