Belimumab is a fully human, IgG1-lambda monoclonal antibody approved for the treatment of active, autoantibody-positive systemic lupus erythematosus (SLE) and active lupus nephritis. It functions as a B-lymphocyte stimulator (BLyS)-specific inhibitor. By binding to and neutralizing the soluble form of the cytokine BLyS (also known as BAFF), it prevents BLyS from interacting with its receptors on B cells. This inhibition reduces the survival and differentiation of autoreactive B cells, leading to a decrease in pathogenic autoantibody production and disease activity.
