Pembrolizumab is a humanized IgG4κ monoclonal antibody that functions as a programmed cell death‑1 (PD‑1) immune checkpoint inhibitor. It binds with high affinity to the PD‑1 receptor on T cells, blocking its interaction with its ligands PD‑L1 and PD‑L2. This blockade reverses PD‑1‑mediated immunosuppression, restoring T‑cell activation and enhancing anti‑tumor immune responses. Pembrolizumab is approved for a broad range of advanced solid tumors and hematologic malignancies, including melanoma, non‑small cell lung cancer (NSCLC), head and neck squamous cell carcinoma, classical Hodgkin lymphoma, gastric cancer, hepatocellular carcinoma (HCC), renal cell carcinoma (RCC), endometrial carcinoma, and many others, both as monotherapy and in combination regimens. It is administered via intravenous infusion.
