Enfortumab vedotin is an antibody-drug conjugate (ADC) approved for the treatment of locally advanced or metastatic urothelial carcinoma. It consists of a fully human monoclonal antibody linked to a potent cytotoxic payload, monomethyl auristatin E (MMAE), via a protease-cleavable linker. Enfortumab vedotin functions as a targeted delivery system for chemotherapy. The antibody component specifically binds to Nectin-4 on cancer cells, leading to ADC internalization. Inside the cell, the linker is cleaved, releasing MMAE, which disrupts microtubule networks, inhibits cell division, and induces programmed cell death.
