Pucotenlimab (also known as HX008 and the proposed international nonproprietary name Pucotenlimab) is a humanized IgG4 monoclonal antibody developed for cancer immunotherapy. It specifically targets the Programmed Cell Death Protein 1 (PD-1) receptor, a key immune checkpoint expressed on activated T-cells and other immune cells. By binding with high affinity to PD-1, pucotenlimab competitively blocks its interaction with its ligands, PD-L1 and PD-L2, which are often overexpressed on tumor cells and within the tumor microenvironment. This blockade releases the inhibitory "brake" on T-cell activity, thereby reversing T-cell exhaustion, restoring and enhancing cytotoxic T-cell function, and promoting an effective anti-tumor immune response against cancer cells.
