Lacnotuzumab (also known as MCS110) is a human monoclonal antibody that targets colony-stimulating factor 1 (CSF-1 or M-CSF). It functions by binding to and neutralizing circulating CSF-1, thereby inhibiting its interaction with the CSF-1 receptor (CSF-1R) on target cells. This blockade aims to deplete tumor-associated macrophages (TAMs), particularly the pro-tumorigenic M2 phenotype, within the tumor microenvironment. By modulating the immune landscape, Lacnotuzumab seeks to enhance anti-tumor immunity and has been investigated in clinical trials for the treatment of various solid tumors, such as tenosynovial giant cell tumor (TGCT) and certain advanced cancers, often in combination with other therapies.
