Tremelimumab is a fully human monoclonal antibody targeting cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), an immune checkpoint receptor. It binds to CTLA-4 on T cells, blocking interaction with its ligands (CD80/CD86) to enhance T cell activation and proliferation. It is approved in combination with durvalumab (anti-PD-L1) for unresectable hepatocellular carcinoma and metastatic non-small cell lung cancer, and is studied in other solid tumors.
