Tilogotamab (GEN-1029) is a humanized IgG1 kappa monoclonal antibody targeting death receptor 5 (DR5, also known as TRAIL-R2 or CD262), a pro-apoptotic receptor in the TNF superfamily. It functions as an agonist, binding to DR5 to induce clustering and activation of the extrinsic apoptotic pathway in tumor cells. It is being developed for the treatment of solid tumors, such as colorectal and pancreatic cancers, where DR5 is often overexpressed.
