名称PTP1B-IN-33
描述PTP1B-IN-33 is a protein tyrosine phosphatase 1B (PTP1B) inhibitor with an IC₅₀ of 2.45 μM against human PTP1B, showing more than 20-fold selectivity over SHP2. It enhances π-alkyl interactions with PTP1B, thereby promoting closure of the WPD loop. PTP1B-IN-33 can be used in studies on cancer, diabetes, and autoimmune deficiency diseases.
体外活性方法： 采用 PTP1B-IN-33处理人源 PTP1B 与 SHP2 PTP 蛋白，检测其结构变化及相互作用。 结果： PTP1B-IN-33 诱导人源 PTP1B 的 WPD 环闭合程度（5.4 Å）显著高于人源 SHP2 PTP（1.2 Å），并通过与磷酸酪氨酸（pTyr）环残基 R47 的强相互作用增强 PTP1B 稳定性 [1]。
