描述LY2928057 is a humanized antibody targeting the iron transporter SLC40A1 and acts as an iron regulator inhibitor, making it suitable for research into cardiovascular diseases. By blocking iron regulator binding and stabilizing the cell surface iron transporter, LY2928057 promotes iron release and utilization, making it applicable to research on anemia-related conditions such as chronic inflammatory anemia.
体外活性LY2928057 是靶向铁转运蛋白的单克隆抗体。LY2928057 结合铁转运蛋白并阻断与铁调素的相互作用，导致铁外流，导致猴子和人类血清铁和转铁蛋白饱和度水平升高，铁调素增加。[1] 方法：选用人肺动脉平滑肌细胞（hPASMCs），LY2928057（1 μg/mL）预处理 1.5 h 进行干预，然后给予铁调素（1 μg/mL）刺激，处理时间2.5 h、24 h、48 h。RT-PCR、Western blot、ELISA、免疫荧光检测铁转运蛋白（ferroportin）、铁调素、CD163、IREB2表达。 结果：LY2928057 可阻断铁调素结合铁转运蛋白导致的其内吞降解，细胞铁蓄积。[3]
体内活性LY2928057（10 - 30 mg/kg ） 给药导致食蟹猴血清铁调素剂量依赖性增加.[2]
