名称Methotrexate α-tert-butyl ester
描述Methotrexate α-tert-butyl ester is a protected analog of the antineoplastic and immunosuppressive drug methotrexate (MTX), in which the β-carboxyl group of the L-glutamate moiety is esterified with a tert-butyl group. Methotrexate α-tert-butyl ester is primarily used in chemical synthesis and prodrug development studies.
体外活性MTX-PVG对HT1080细胞表现出活性，但OtBu-MTX-PVG衍生物(Methotrexate α-tert-butyl ester)的活性较低，这表明保护MTX的α-羧基会导致体外活性降低。[1]
体内活性在HT1080荷瘤小鼠模型中，Methotrexate α-tert-butyl ester显著抑制了肿瘤生长。这一发现表明，通过保护甲氨蝶呤的α-羧基，可在不影响体内抗肿瘤功效的前提下，延长类似树枝状聚合物偶联物的循环半衰期并减少肝脏蓄积。[1]
