名称G721-0282
描述G721-0282 is an orally active inhibitor of CHI3L1 that reduces the expression of inflammatory proteins and cytokines. It suppresses the activation of the NF-κB signaling pathway, inhibits neuroinflammation, and decreases anxiety-related behaviors. Additionally, G721-0282 significantly curtails the proliferation of osteosarcoma (OS) cells by inhibiting the STAT3 signaling pathway and induces apoptosis in OS cells by upregulating pro-apoptotic protein levels while downregulating anti-apoptotic protein levels. This compound is useful in the study of neuroinflammatory diseases and cancer.
体外活性G721-0282 (5-20 μM) 有效抑制 BV-2 细胞中 LPS 诱导的神经炎症，涉及 NO 的产生、促炎细胞因子表达以及炎症蛋白水平。使用 G721-0282 (20 μM) 在 BV-2 细胞中依赖 CHI3L1 发挥抗炎效应。它在 (10-50 μM, 24-72 小时) 能以剂量和时间依赖的方式抑制 MG63 和 U2OS 细胞增殖。G721-0282 (0-50 μM, 24 小时) 通过降低抗凋亡蛋白和增加促凋亡蛋白水平来阻断细胞周期并诱导 MG63 和 U2OS 细胞的凋亡。此外，G721-0282 在 (0-50 μM, 12-48 小时) 通过抑制 STAT3 信号路径显著降低 MG63 和 U2OS 细胞的迁移及侵袭能力。使用 G721-0282 长达 (2-4 周) 抑制 OS 细胞在软琼脂中的集落形成，并在 (0-50 μM, 24 小时) 内减少 MMP2 和 MMP9 的水平。
体内活性G721-0282（2.5 和 5 mg/kg；口服；每周两次；持续 4 周）显著减少了雄性 BALB/c 小鼠中由慢性不可预测轻度应激 (CUMS) 所引起的焦虑样行为，并抑制神经炎症。G721-0282（18.9 mg/kg；腹腔注射；每周两次；持续 35 天）在携带 MG63 肿瘤的雄性 BALB/c 无胸腺裸鼠中展示出显著的抗肿瘤活性。
