名称AR antagonist 15
描述AR antagonist 15 is an orally active androgen receptor (AR) antagonist with an IC50 of 97 nM for ART787A. It impairs AR nuclear translocation, hinders AR homodimerization, and inhibits AR-regulated gene transcription by competitively binding to the ligand-binding pocket. AR antagonist 15 significantly reduces prostate-specific antigen (PSA) levels and induces apoptosis by decreasing the expression of apoptosis-related proteins. This compound is useful for prostate cancer research.
体外活性AR antagonist 15 (Compound LT16) 对雄激素受体 (AR) 及其突变体表现出强拮抗活性，包括 AR T787A (IC 50 = 97 nM)、AR F877L (IC 50 = 200 nM)、AR F877L/T787A (IC 50 = 260 nM)、AR H875Y/T787A (IC 50 = 200 nM)、AR W742C (IC 50 = 510 nM) 和 AR T787G (IC 50 = 555 nM)。通过剂量依赖性作用，AR antagonist 15 能够降低前列腺特异性抗原 (PSA) 的分泌，IC 50 为 0.16 μM，并促使 AR 在48小时内从细胞质转移至细胞核。AR antagonist 15 在0.1-10 μM浓度下可于48小时内下调 LNCaP 细胞中 AR 调控基因的 mRNA 水平，并显著降低与凋亡路径有关的重要蛋白的 mRNA 水平。此外，在0.01-100 μM 浓度下，3到5天内显著抑制 AR 阳性细胞的增殖，在10 μM下，48小时内诱导 LNCaP 细胞发生 G0/G1 细胞周期阻滞。在0.5-5 μM浓度下，AR antagonist 15 能在14天内抑制 LNCaP 细胞的生长并减少细胞集落数量。
体内活性AR antagonist 15 以 50 mg/kg 剂量通过 i.g. 方式每日一次给予雄性 Balb/C nude 前列腺癌小鼠，持续三周，显著减少肿瘤体积并降低血清 PSA 水平。
