名称Cyanine 7.5 maleimide chloride
描述Cy7.5 maleimide chloride is a type of CY dye, where CY stands for the cyanine class of compounds, consisting of two nitrogen atoms linked by an odd number of methine units. Cyanine compounds are known for their long wavelengths, adjustable absorption and emission, high extinction coefficients, good water solubility, and relatively simple synthesis. CY dyes are frequently used to label proteins, antibodies, and small molecular compounds. For antibody labeling, the dyes can be conjugated through a straightforward mixing reaction, making them a useful reference for protein and antibody labeling methods.
体外活性蛋白准备：为确保最佳的标记效果，请将蛋白（抗体）浓度配置为2 mg/mL。1) 当蛋白溶液的pH值低于8.0时，需使用1 M 碳酸氢钠调整至8.5±0.5。2) 若浓度低于2 mg/mL，则标记效率显著降低，建议最终蛋白浓度为2-10 mg/mL。3) 蛋白应置于无伯胺和铵离子的缓冲液中，以免影响标记效率。染料准备：CY染料用无水DMSO稀释成10 mM储备溶液，并通过玻璃管或旋涡充分混合。注意，将CY储备液分装后，于-20℃或-80℃避光保存。染料用量：标记所需的CY染料量由蛋白量决定，CY染料与蛋白的最佳摩尔比为10左右。例如：若需标记500 μL 2 mg/mL的IgG (MW=150,000)，用100 μL DMSO溶解1 mg CY染料，则需3.95 μL CY，具体计算如下（以CY3-NHS ester为例）：1) mmol (IgG) = 6.7×10^-6 mmol 2) mmol (CY3-NHS ester) = 6.7×10^-5 mmol 3) μL (CY3-NHS ester) = 3.95 μL (CY3-NHS ester)。使用方法：标记反应：1) 将新配制的10 mg/mL CY染料缓慢加入0.5 mL蛋白样品溶液，轻轻摇匀混合后短暂离心。2) 在避光处室温摇晃孵育60分钟，每10–15分钟轻轻颠倒以提高标记效率。蛋白纯化脱盐：使用SepHadex G-25柱。1) 准备SepHadex G-25柱。2) 将反应混合物装入柱顶部。3) 样品到达树脂下方时，立即加入PBS (pH 7.2-7.4)。4) 继续加入PBS至柱纯化完成，收集含有染料-蛋白质缀合物的组分。 以上信息均来源于已发表文献，具体实验方案请根据实际研究需求进行适当调整。
