名称Geiparvarin
描述Geiparvarin is an anticancer agent that acts as an inhibitor of MAO-B (pIC50= 6.84 μM). It exerts its antitumor effects by downregulating COX2 expression and inhibiting angiogenesis. Geiparvarin halts the cell cycle at the G1 phase and induces apoptosis in cancer cells. Additionally, it exhibits antimicrotubule activity (Microtubule/Tubulin) and disrupts the cytoskeleton, contributing to its antiproliferative properties. Geiparvarin is of research interest for its potential effects on lung cancer, leukemia, and breast cancer.
体外活性Geiparvarin (Compound 8) 在72小时和48小时内对多种人类肿瘤细胞系的增殖具有抑制作用 (IC 50: 5.7 μM用于SHSY5Y, 6.3 μM用于HL-60, 9.2 μM用于K562, 9.8 μM用于HT-1080, 11.5 μM用于A-549)。 Geiparvarin (8 μM, 24-72小时) 阻滞HL-60细胞周期于G1期，并诱导非caspase依赖性凋亡。 在72小时内，Geiparvarin 对耐药细胞系表现出完全抑制，且不受介导抗肿瘤药物外排的转运蛋白的影响。 Geiparvarin (1 μM, 24小时) 抑制HOS和143B细胞的侵袭能力。 在HOS和143B细胞中，Geiparvarin (0.5-2 μM, 24-48小时) 激活凋亡通路，抑制COX2及其下游的血管生成通路，并诱导caspase依赖性凋亡。 在骨肉瘤细胞中，Geiparvarin 通过COX2下调发挥其抗肿瘤作用，且COX2过表达逆转其对增殖和侵袭的抑制。 Geiparvarin (10 μM, 24小时) 在Balb/c 3T3成纤维细胞中破坏细胞骨架结构，特别是中间丝。 Geiparvarin (0.01-10 μM) 在鼠肝线粒体提取的MAO-B中，随着浓度的增加，抑制率显著上升，pIC 50 = 6.84 (IC 50 = 1.45 μM)。
体内活性Geiparvarin (Compound 8) （0.5 mg/kg, i.p, 每三天或每天持续数周）在裸鼠的143B/9901细胞异种移植瘤模型中有效抑制肿瘤生长和肺部转移，并且无显著毒性。
