名称3-(2-Hydroxyphenyl)-2-propenal
描述3-(2-Hydroxyphenyl)-2-propenal is a natural phenylpropanal isolated from the bark of the cassia cinnamon (Cinnamomum cassia). 3-(2-Hydroxyphenyl)-2-propenal inhibits the proliferation of various tumor cells and induces apoptosis by directly binding to STAT3 and inhibiting its phosphorylation at Tyr705 and nuclear translocation, while simultaneously regulating the expression of downstream target genes by inhibiting the Wnt/β-catenin signaling pathway. (E)-2-Hydroxycinnamaldehyde possesses antitumor activity and can be used in tumor research.
体外活性方法：DU145 前列腺癌细胞加入 5~40 μM 3-(2-Hydroxyphenyl)-2-propenal 处理 24~48 h，WST-1 法检测细胞增殖，Western blot 检测 STAT3 磷酸化水平。 结果：DU145 细胞增殖 GI₅₀=20 μM；20 μM 3-(2-Hydroxyphenyl)-2-propenal 处理 30 min 即显著抑制 STAT3 Tyr705 磷酸化，12 h 抑制率 > 70%，不改变总 STAT3 蛋白水平。[1] 方法：20 μM 3-(2-Hydroxyphenyl)-2-propenal 处理 DU145 细胞 24~48 h，Western blot 检测 STAT3 靶基因表达；流式细胞术检测细胞周期分布。 结果：3-(2-Hydroxyphenyl)-2-propenal 显著下调细胞周期基因（cyclin D1、cyclin A）和抗凋亡基因（Bcl-2、Bcl-xL、Mcl-1、survivin）；6 h 开始出现 G0/G1 期阻滞，24 h 后凋亡细胞（sub-G1 峰）比例显著升高。[1]
体内活性方法：6 周龄雌性 BALB/c 裸鼠皮下接种 1×10⁶ DU145 细胞，随机分组后口服给药：3-(2-Hydroxyphenyl)-2-propenal 50 mg/kg，每周 5 次，连续 25 天；每周 2 次测量肿瘤体积与小鼠体重，终点处死小鼠称量肿瘤重量。 结果：3-(2-Hydroxyphenyl)-2-propenal 显著抑制肿瘤生长，与对照组相比，肿瘤体积减少 48.4%，肿瘤重量减少 49.5%；给药期间小鼠体重无明显下降，无明显毒性反应。[1]
