Feladilimab (development code GSK-3359609) is an investigational, humanized monoclonal IgG4 antibody developed for cancer immunotherapy. It functions as an agonist of the Inducible T-cell Co-Stimulator (ICOS) receptor. By binding to ICOS on the surface of activated T cells, feladilimab mimics the action of its natural ligand (ICOS-L), delivering a potent co-stimulatory signal. This signal enhances T-cell proliferation, survival, cytokine production (particularly IL-10 and IFN-γ), and supports the function of effector T cells and T follicular helper (Tfh) cells. The goal is to amplify the anti-tumor immune response, especially when combined with other immunotherapies that initially activate T cells, such as PD-1/PD-L1 checkpoint inhibitors.
