Tegoprubart is a humanized IgG1 kappa monoclonal antibody targeting CD40 ligand (CD40L, also known as CD154), a transmembrane protein primarily expressed on activated T cells. It binds to CD40L, blocking its interaction with CD40 on antigen-presenting cells (e.g., B cells, dendritic cells) and endothelial cells. This inhibition suppresses T cell-dependent immune activation, B cell maturation, and inflammatory signaling. It is being developed for the treatment of autoimmune diseases such as systemic lupus erythematosus (SLE) and transplant rejection.
