名称G-418
描述G-418 (Geneticin) is an aminoglycoside antibiotic structurally similar to gentamicin, exhibiting cytotoxicity against both eukaryotic and prokaryotic cells. The mechanism of action of G-418 (Geneticin) involves interference with protein synthesis, and it is commonly used in molecular biology for screening and maintaining cell lines stably transfected with neomycin resistance genes.
体外活性方法：G-418 用于筛选表达人GIP或胰高血糖素受体构建体的重组BHK-570和CHO-K1细胞，并监测细胞生长和代谢。 结果：持续G418筛选产生细胞系依赖的代谢负担，并改变生长和营养物代谢；移除G418后重组蛋白表达仍保持稳定。[1] 方法：G-418 与其他氨基糖苷类加入哺乳动物细胞，并检测糖基磷脂酰肌醇锚定蛋白的释放。 结果：G418促进GPI锚定蛋白从细胞表面释放，表明其除抗生素筛选外还具有直接细胞作用。[2]
