In a sealed tube, 3-chloropyrazin-2-amine (10 g, 77 mmol) and morpholine (40 mL, 463 mmol) were mixed and the reaction was heated at 120 °C for 12 hours. After completion of the reaction, the reaction mixture was diluted with ethyl acetate (200 mL), washed sequentially with water and brine, dried over anhydrous sodium sulfate, filtered, and concentrated under reduced pressure. The concentrated residue was recrystallized by ethyl acetate and filtered to obtain 2-amino-3-morpholin-4-ylpyrazine (white solid, 7.45 g). The filtrate was further concentrated and purified by silica gel column chromatography (60-90 °C petroleum ether: ethyl acetate = 1:1) to give a second batch of 2-amino-3-morpholin-4-ylpyrazine (white solid, 5.38 g).
