To a mixed solution of 2-aminoethane-1-thiol (4.5 g, 40 mmol, 1.0 eq.) and potassium carbonate (K2CO3, 5.5 g, 40 mmol, 1.0 eq.) in ethanol (EtOH, 80 mL) was slowly added ethyl chloroacetate (40 mmol, 1.0 eq.). The reaction mixture was stirred at room temperature for 2 hours, followed by warming to 75 °C and continued stirring for 24 hours. Upon completion of the reaction, the insoluble material was removed by filtration and all volatile solvents were removed by rotary evaporator under reduced pressure. The crude product was purified by fast column chromatography (silica gel, ethyl acetate as eluent) to give the final thiomorpholin-3-one as a white solid in 18% yield. | [References]
