In an oven-dried Schlenck reaction tube, the corresponding 4-nitroaniline (0.50 mmol) and salicylic acid (6.9 mg, 0.05 mmol) were added. The system was evacuated and purged with argon (three times), and then the corresponding enol acetate (5 mmol), solvent MeCN (1.65 mL), and H2O (0.85 mL) were added to the reaction system. The resulting reaction mixture was stirred vigorously until a homogeneous solution was obtained (approximately 1–5 min). Then, tert-butyl nitrous acid (100 μL, 86 mg, 0.75 mmol) was added to the reaction system (the solution turned orange after a few minutes). The reaction mixture was stirred for 3 h, and the reaction temperature was maintained at around 20 °C using an external water bath. After the reaction was complete, water was added to the reaction mixture, and the reaction mixture was extracted three times with ethyl acetate (3 × 15 mL). The organic layer was collected and dried over anhydrous MgSO4, the desiccant was removed by filtration, and the resulting filtrate was concentrated under vacuum. The resulting residue was purified by silica gel column chromatography (n-hexane/ethyl acetate 95:05) to obtain 4-NITROPHENYLACETONE. Figure 4-Nitrophenylacetone Synthetic Route |
