Ethyl 2-((Tetrahydro-2H-pyran-2-yl)oxy)acetate has been used as a reactant for the sysnthesis of 3-phenylpyrazolo[1,5-a]pyrimidine as a potent corticotropin-releasing factor-1 antagonist. | [Synthesis]
The general procedure for the synthesis of ethyl 2-((tetrahydro-2H-pyran-2-yl)oxy)acetate from 3,4-dihydro-2H-pyran and ethyl ethanoate was as follows: to a solution of toluene (100 mL) containing ethyl ethanoate (10 g, 0.0961 mol), 3,4-dihydro-2H-pyran (8 g, 0.096 mol) was added followed by catalytic amounts of p Toluenesulfonic acid (pTSA, 30 mg). The reaction mixture was stirred at 25°C for 1 hour. After completion of the reaction, the reaction mixture was washed sequentially with water and brine solution. The organic layer was separated, dried over anhydrous sodium sulfate (Na2SO4), filtered and concentrated under reduced pressure to afford 18 g (99% yield) of ethyl 2-((tetrahydro-2H-pyran-2-yl)oxy)acetate as a colorless liquid. | [References]
