4-tert-Butylbenzenesulfonamide is an intermediate as well as a degradation impurity of the endothelin receptor antagonist Bosentan (B675900). | [Synthesis]
A solution of 4-(tert-butyl)benzenesulfonyl chloride (2.30 g, 10.0 mmol) in dichloromethane (100 mL) was added to a 500 mL round bottom flask at 0 °C, followed by the slow addition of concentrated ammonia (50 mL, 100 mmol, 10 equiv). The reaction mixture was gradually warmed to room temperature and stirred continuously for 20 hours. Upon completion of the reaction, the solvent was removed by evaporation under reduced pressure and the resulting slurry was filtered through a Brinell funnel to afford 1.60 g (75% yield) of 4-(tert-butyl)benzenesulfonamide (Compound A) as a white solid. | [References]
