以2-氯-5-氰基吡啶和(R)-(-)-2-甲基哌嗪为原料合成(R)-6-(3-甲基哌嗪-1-基)烟腈的一般步骤如下:向含有6-氯吡啶-3-甲腈(10g,0.06mol)和(2R)-2-甲基哌嗪(6.35g,0.06mol)的乙腈(80mL)溶液中加入K2CO3(12.0g,0.09mol)。在室温下,将反应混合物加热至60℃并搅拌2小时(通过TLC监测反应完成)。待反应冷却至室温后,用水(150mL)淬灭反应,随后用EtOAc(3×80mL)进行萃取。合并有机相,用无水Na2SO4干燥,减压浓缩。残余物通过柱色谱法(100-200目硅胶,5% MeOH-DCM作为洗脱剂)纯化,得到目标产物(R)-6-(3-甲基哌嗪-1-基)烟腈(10.0g,收率69%)。
