以环己酮和氰乙酸甲酯为原料,通过标准Gewald反应合成2-氨基-4,5,6,7-四氢苯并[b]噻吩-3-羧酸甲酯(化合物1)。具体步骤如下:向环己酮(4.91g,0.05mol)、氰乙酸甲酯(4.95g,0.05mol)和硫(1.92g,0.06mol)在35mL乙醇中的搅拌溶液中,缓慢滴加吗啉(4.4g,0.05mol)。滴加完毕后,将反应混合物回流3小时。反应完成后,冷却至室温,过滤收集沉淀物。通过乙醇重结晶纯化,得到浅黄色粉末状产物(8.6g,产率82%)。产物的熔点为128.2-129.4℃(文献值:128-130℃),质谱(GC-MS)分析显示m/z为211 [M + H]+,碎片离子峰为179、151、125、91、77、65、53。
